| Treatment Conditions: | supplier: Plants from pathogen-free Solanum tuberosum var. Shepody, clone 1756, nuclear stock were grown in a screenhouse under natural conditions. Stolons and mature tubers were used for total RNA isolations. Two hundred and fifty nanograms of each sample were pooled and used for full length 1st strand cDNA synthesis with a modified SMART cDNA kit (BD Biosciences), in which the kit CDS III primer was redesigned to contain an XhoI site. This was followed by PCR using the redesigned CDS III primer and the kit 5â PCR primer. PCR products were blunted, and ligated to EcoRI adapters (Invitrogen), followed by XhoI digestion. The double stranded, digested cDNA was gel-purified, and the fraction above 650 bp was collected and ligated with the vector. |
