Comments | This library targets genes expressed during the wound-healing process of periderm formation. Potatoes were peeled well away from the surface and sliced into 1-2cm thick rounds. They were then placed on toothpicks, which were placed into a Stytrofoam lid. This was covered with saran wrap which was punctured several times. This enabled air to flow on all sides of the potato round. They were stored in an incubator in the dark at 22C. No evidence of infection was present on any batches of collected tissue. The periderm collected was separated into surface periderm ( a dry, flaky, scab-like layer on the surface) and subperiderm (a browning, moist layer 1-2 mm below the surface of the periderm). Both periderm and subperiderm were collected at 24 hours, 48 hours, 5 days, and 7 days. Tissue was frozen in liquid nitrogen immediately upon collection. Later, the mRNA was pooled from each. |
